Best peptides for growth hormone, ranked by the human evidence
8 compounds are sold for raising growth hormone, muscle and fat loss. We rank them by how strong the evidence in people is, not by popularity. 1 of the 8 is FDA-approved for some use; the rest are not.
Tesamorelin, sermorelin, MK-677, GHRP-2, GHRP-6, hexarelin, CJC-1295 (in its long-acting DAC form) and ipamorelin all raise growth hormone in people, but a higher hormone level is not proof of fat loss or muscle gain123456789. In trials in people with an illness, tesamorelin cut deep belly fat in adults with HIV without changing their weight, and sermorelin sped up growth in children who make too little growth hormone, though growth hormone itself did as well or better1011121314. MK-677 did not slow Alzheimer’s disease in 563 people, and CJC-1295’s only patient trial was stopped in 2006, and we found no published results15161718. GHRP-2 is approved in Japan only as a one-dose test for growth hormone deficiency, and its nasal spray did not help short children grow in the one placebo-controlled trial192021. GHRP-6’s largest trials paired it with another drug in stroke, and the larger of the two missed its main goal2223. Hexarelin’s effect on growth hormone faded with daily use, and in 12 older adults given it for 16 weeks, with no comparison group, body fat and lean mass did not change significantly2425.
By our reading, US pharmacies can probably make sermorelin on prescription, because FDA lists it as a component of an approved drug, but not tesamorelin, which FDA classes as a biologic, a kind of drug the compounding rules exclude26272829. They may not make CJC-1295, ipamorelin or MK-677, which FDA’s advisers voted against allowing in October and December 2024303132. Nor, by our reading, may they make GHRP-2, GHRP-6 or hexarelin: FDA puts the first two in a category it says it may act against pharmacies for using, and hexarelin is on none of its compounding lists2730. FDA also says MK-677 cannot be sold as a dietary supplement33. On 24 August 2026, FDA told online sellers that their tesamorelin products, including blends with ipamorelin, were unapproved new drugs3435. Many samples sent for testing fail: in tests up to September 2026 by Finnrick, a testing service, 124 of its latest 200 CJC-1295 samples failed, as did 52 of its latest 200 ipamorelin samples, 83 of the 191 completed tests among its latest 200 for tesamorelin and 27 of 48 completed sermorelin tests36373839. People choose which samples to send, so the results are not a random survey, and Finnrick had listed no MK-677 tests as of 25 September 2026, or any tests of GHRP-2, GHRP-6 or hexarelin as of 7 October 2026363738394041424344.

Tesamorelin
Tesamorelin boosts growth hormone and is FDA-approved only for excess belly fat in adults with HIV1345. Other uses have little evidence946, and vials sold online often fail tests39.
Does it work for reducing deep belly fat? In two large HIV trials, deep belly fat fell about 15% more than on placebo, then returned after stopping144748. Outside HIV, a 12-month trial lowered it, in people with obesity and low growth hormone output49.
-
Sermorelin
Sermorelin is a lab-made piece of a brain hormone that tells the body to release growth hormone. Once approved in the US for children, it was discontinued in 2008, and its studies in adults are small and short5051.
Does it work for anti-aging? In a 6-month trial against placebo in 89 older adults, some thinking tests improved52. The team’s other reports found less body fat but no better sleep, and said routine use was not yet supported5153.
-
MK-677 (ibutamoren)
MK-677 is an experimental pill, not a peptide, that makes the body release more growth hormone54. In older adults it added lean mass but not strength and raised blood sugar55; a hip-fracture trial stopped early over a heart failure signal56.
Does it work for building muscle and lean mass? In older adults, a year of daily pills added about 1 kg of lean mass but did not improve strength or function55. In obese men, lean mass rose but body fat did not fall57.
-
GHRP-2 (pralmorelin)
GHRP-2 is a lab-made peptide that makes the body release growth hormone19. Japan approved it in 2004 as a one-dose test for growth hormone deficiency1920. We found no trial of it for muscle gain or fat loss258.
Does it work for muscle gain and fat loss? It raises growth hormone and appetite359. We found no trial of muscle gain or fat loss; a chart review of men seeking both measured only a blood marker of growth hormone58.
-
GHRP-6
GHRP-6 was one of the first lab-made peptides shown to make the body release growth hormone24. It is not approved in the US60, and we found no trial of it for muscle gain or fat loss2.
Does it work for muscle gain and fat loss? It raises growth hormone and the stress hormones ACTH and cortisol in healthy men61. We found no trial of muscle gain or fat loss; its largest trials paired it with another drug for stroke2223.
-
Hexarelin
Hexarelin, a lab-made peptide that makes the body release growth hormone5, briefly made the heart pump harder in small one-dose studies6263. It is not approved in the US, and we found no controlled trial of it for muscle gain264.
Does it work for muscle gain and fat loss? It raises growth hormone in healthy adults5. In 12 older adults injected for 16 weeks, body fat and lean mass did not change significantly, but there was no comparison group25.
-
Ipamorelin
Ipamorelin is a lab-made peptide that triggers short bursts of growth hormone6. Its only published patient trial found no clear benefit, and we found no study testing it in people for fat loss, muscle or sleep656667.
Does it work for fat loss, muscle and sleep? An IV dose triggers a short burst of growth hormone6, but we found no study in people of fat loss, muscle or sleep6667. Its only published patient trial, after bowel surgery, found no clear benefit65.
-
CJC-1295
CJC-1295 is a lab-made version of a brain hormone that makes the body release growth hormone15. It is not approved in the US or Europe, and we found no published study of fat loss, muscle or healing in people1217.
Does it work for fat loss, muscle and healing? Published human studies used only the ‘with DAC’ form, and one dose raised growth hormone for 6 days or more115. We found no published study of fat loss, muscle or healing in people1217.
How we ranked them
Compounds that trials show work for this goal come first, then those with mixed results, then those with no controlled human trial for this goal that has public results, and last those that trials found do not work. Within each group, the stronger the human evidence (tier A to E), the higher the place. Ties go to compounds FDA has approved as a medicine, then to those approved only in other countries, then to those whose US approval was withdrawn, then to peptides over non-peptide drugs, then to the winner of a trial that compared the two directly. Popularity, price and how a product is marketed play no part. A high place is not a recommendation to use anything.

Questions people ask
What is the best peptide for growth hormone?
There is no proven best. For muscle, fat loss or aging in healthy adults, the trials we found of tesamorelin, sermorelin, MK-677, CJC-1295 and ipamorelin are small or short, and for some of them we found none1946515566. We found no controlled trial that tested GHRP-2, GHRP-6 or hexarelin for muscle gain or fat loss2. Tesamorelin is FDA-approved, but only for belly fat in adults with HIV, and its label says it is not for weight loss1345.
Do growth hormone peptides build muscle?
Not clearly. In MK-677’s longest trial, a year of daily pills in older adults raised fat-free mass, which includes muscle and body water, by 1.1 kg against a 0.5 kg fall on placebo, but strength and physical function did not change, though the trial was too small to test them well55. In 11 older men, 6 weeks of nightly sermorelin left muscle on body scans unchanged, though 2 of 6 strength measures improved, and we found no published study of CJC-1295 or ipamorelin for muscle in people1217666768. In 12 older adults, 16 weeks of hexarelin injections left lean mass and body fat unchanged, though that study had no comparison group25.
Are growth hormone peptides safe?
Long-term safety in healthy adults is unknown: the adult sermorelin trials we found lasted 6 months or less, and we found no long-term data for CJC-1295 or ipamorelin151751526667. In tesamorelin’s trials, 5% of treated patients reached a diabetes-range HbA1c, a long-term blood sugar measure, against 1% on placebo, and a growth factor rose above normal in 47% of treated patients, and well above in 36%, which may raise cancer risk1369. MK-677 raised blood sugar in older adults, and a hip-fracture trial of it was stopped early after heart failure in 4 patients on the drug against 1 on placebo545556. GHRP-2, GHRP-6 and hexarelin also raise the stress hormones ACTH and cortisol617071.
GHRP-2 vs GHRP-6 vs hexarelin: what is the difference?
All three are lab-made chains of six amino acids that act like ghrelin, the hunger hormone, to release growth hormone272. GHRP-6 came first, in 1984; GHRP-2 was later developed as a stronger version and is approved in Japan as a one-dose test, and hexarelin is GHRP-6 with one tryptophan swapped for a more stable form19707374. We found no controlled trial that tested any of them for muscle gain or fat loss2.
Will growth hormone peptides fail a drug test?
In anti-doping tests, they can. Tesamorelin, sermorelin, MK-677, GHRP-2, GHRP-6, hexarelin, CJC-1295 and ipamorelin are all banned in sport at all times75. In February 2026, the US Anti-Doping Agency announced a 3-year ban, starting in December 2025, for a masters weightlifter whose urine sample tested positive for ipamorelin, an ipamorelin breakdown product and ibutamoren (MK-677)76.
